Histology

The BIOSCAR histology core facility specialises in the processing and analysis of mineralised tissues.
It is open to various teams on site as well as to external teams, whether from institutions or the private sector. All equipment is available for use, either on a loan basis with staff training or as a service provided on request following a project assessment. The platform is committed to providing a high-quality service combined with personalised support throughout the implementation of the project.

/ Expertises

1. Non-decalcified hard tissues study (embedding in methyl methacrylate resin) :

  • Quantification of static bone parameters (labelling or staining of osteoblasts, osteoclasts and osteocytes)
  • Quantification of dynamic formation parameters
  • Analysis of the bone matrix and its state of mineralisation
  • Preparation of thick sections, for example for the analysis of microcracks

2. Decalcified hard tissues or soft tissues study (embedding in paraffin or OCT) :

  • Quantification of cell proliferation/survival parameters (apoptosis of bone cells, proliferation)
  • Study of vascularisation and the medullary microenvironment via immunolabelling
  • Characterisation of bone cells and their differentiation states via immunolabelling
  • Histology can also be combined with imaging analysis offered on our other dedicated platform, including for measurements of bone density and morphology

/ Equipment

Sample preparation

  • 1 paraffin embedding machine
  • 1 MM France EC500 paraffin embedding platform

Sample sectioning

  • 2 resin microtomes (Leica SM2500 and Leica SM2500S)
  • 1 ThermoScientific NX70 cryostat
  • 2 paraffin microtomes (Leica RM2255 and Leica Autocut)
  • 1 Q ATM QPOL 250 M1 polisher
  • 1 Buehler Low Speed Saw isometer

Image analysis

  • 2 optical microscopy stations coupled with Microvision Instruments image analysers (Nikon Optiphot2 and Nikon Eclipse 50 with polarised light)
  • 1 optical microscopy station with a motorised stage coupled with a Microvision Instruments touchscreen and stylus image analyser (Zeiss AxioImager)
  • 1 fluorescence microscope with optical sectioning via Zeiss Apotome structured illumination (DAPI, FITC, dsRed, Cy5)
  • 1 Zeiss LSM800 confocal microscope (DAPI, FITC, Rhodamine filter cube) with motorised stage and 405, 488, 561 and 640 nm lasers
